Service Description

We are a certified Oxford Nanopore Technologies provider with years of experience helping clients use Third-Generation Sequencing techniques to achieve their research goals.

Oxford Nanopore Technologies developed unique platforms that allow for long reads of contiguous DNA strands. These platforms use flow cells with pore and membrane technology to read strands of DNA by base associated electrical signal.

We operate a Nanopore GridION Mk1 which runs 5 flow cells at a time and generating upwards of 10Gb of data per flow cell. We have recently purchased a PromethION 24, capable of running 24 flow cells at a time and generating on average, more than 100 Gb of data per flow cell. PromethION services are currently in evaluation.

Advantages

Oxford Nanopore Technologies library prep kits allow us to sequence native DNA, eliminating PCR bias from the data. Native DNA in sequencing maintains strand methylation status. The pore-based flow cells allow for superior reading through long repeat regions compared to traditional next-generation sequencing. These long reads are significant for detecting structural variants throughout the genome. Past projects have consistently generated strands upwards of 100kb (kilobases).

Recommendations

To best take advantage of this sequencing method, we recommend that you submit very high-quality DNA. We offer High Molecular Weight Extraction services. We have successfully extracted High Molecular and Ultra High Molecular Weight DNA from a wide variety of microorganisms, plants, and animals.

To complement our Oxford Nanopore Technologies services, we offer PacBio HiFi and we have had great success with projects that use this service performed in tandem with Oxford Nanopore Technologies to fully elucidate long stretches of genomes.

Many projects require special upstream attention and handling before sequencing on the GridION or PromethION; we would love to meet with you and go over your project plan so we can best assist with offering Oxford Nanopore Technologies services and create a Project Summary Plan to meet your goals.

New Lab or Customer Access

We MUST have this information before we can proceed with your request.

Adding a new lab:

    • Name, address, phone number, UW NetID and email of the Principal Investigator (PI) or Primary scientific contact person.
    • Name, address, phone number, FAX number and email of he financial contact person.
    • Name, UW NetID, phone number and email of all individuals who will be using our service.
    • Payment information: For campus users this should be a direct charge number, e.g. a 144 or 133 prefixed account (please send the whole string of characters). For off campus users you can set up a PO or we can send the financial contact person a bill — your choice. Non-UW system users are charged an additional institutional overhead.

Adding a member to an existing lab:

    • Name of the PI whose lab you are joining.
    • Your name, UW NetID, phone number and email.

Sample Submission

Please contact nextgen-seq@biotech.wisc.edu to arrange all sample submissions. Samples and submission forms must be submitted before a project can be entered into our queue.

Submitting Samples/ Shipping Instructions

After setting up a customer account, filling out submission form, and reviewing submission guidelines, you can now submit or send samples. *We require customers to submit electronic Submission Forms!*

In Person Submission

Please give advance notice of submission date and time so staff can be prepared to receive samples. Schedule a time for drop off with a staff member at nextgen-seq@biotech.wisc.edu
